Strategi Modifikasi Permukaan dalam Penghantaran Oral Biofarmasetika: Inovasi untuk Meningkatkan Bioavailabilitas Insulin dan Vaksin

Obat biofarmasetika adalah produk obat yang diproduksi atau disintesis dari sumber biologis seperti insulin, vaksin, dan makromolekul lain yang memiliki keterbatasan dalam penghantaran oral. Narrative review ini bertujuan mengevaluasi strategi aktivasi permukaan pada sistem penghantaran obat biofarmasetika oral untuk meningkatkan bioavailabilitas dengan mengatasi rintangan biologis di saluran pencernaan. Metode yang digunakan adalah narrative review dengan menganalisis artikel ilmiah dari PubMed, ScienceDirect, dan Google Scholar yang dipublikasikan dalam 13 tahun terakhir menggunakan kata kunci terkait “surface activation oral delivery”, “surface modification”, “oral delivery”, “surface activation vaccine oral delivery”, “oral vaccine”, “oral insuline”. Hasil review menunjukkan bahwa modifikasi permukaan nanopartikel menggunakan ligan penarget spesifik, pelapisan mucoadhesive, polimer responsif pH, dan teknologi biomimetik efektif meningkatkan stabilitas, pelepasan terkendali, penyerapan intestinal pada insulin, dan respons imun vaksin oral. Kesimpulannya, aktivasi permukaan merupakan pendekatan yang menjanjikan untuk pengembangan formulasi insulin dan vaksin yang lebih efektif dan aman dengan bioavailabilitas dan efikasi klinis yang optimal.
